Yes. Again, "prevent all damage from such and such source(s) would deal this turn" still works if the source(s) is/are dealing damage multiple separated times in the same turn.

That's correct, once Sphere's ability has resolved, all damage the source you chose (here, Manabarbs) would deal to you this turn is prevented. Doesn't matter if the damage comes from a separate trigger, the source is still Manabarbs.
