1) No. Both triggers go on the stack at the same time, and the source of one leaving the battlefield will not stop it from resolving.
2) The Gift will return the creature during that end step, but the aura itself will return at the begining of the next turn's end step since the beginning of this end step has already passed, so the delayed trigger has to wait for the next occurence of its trigger condition. However, if you control the Portal, you control its trigger and you can order it and the delayed trigger from the Gift during the next turn's end step in such a way, that the aura returns before any sacrificing is due.

Of course, Kynaios has to actually die for the Gift to trigger, which means, that you cannot put it into the command zone but have to let it hit the graveyard when you sacrifice it. The term "dies" means to be put into the graveyard from the battlefield, and that event has to actually occur to trigger the Gift.

"Miss" their trigger? If that creature is on the battlefield at the beginning of your end step, that ability does trigger, and it would take something like Stifle or you leaving the game to keep it from resolving.

The Gift of Immortality won't be brought back by its delayed trigger until the next end step (presumably your opponent's).
513.2. If a permanent with an ability that triggers “at the beginning of the end step” enters the
battlefield during this step, that ability won’t trigger until the next turn’s end step. Likewise, if a
delayed triggered ability that triggers “at the beginning of the next end step” is created during this
step, that ability won’t trigger until the next turn’s end step. In other words, the step doesn’t “back
up” so those abilities can go on the stack. This rule applies only to triggered abilities; it doesn’t
apply to continuous effects whose durations say “until end of turn” or “this turn.” (See rule 514,
“Cleanup Step.”)

Your end step begins. This triggers the abilities of Kynaios and Tiro of Meletis and Possessed Portal.
You would get priority, but there are two triggered abilities to put on the stack. A) You put "At the beginning of your end step, draw a card. Each player may put a land card from his or her hand onto the battlefield, then each opponent who didn't draws a card." on the stack. B) You put "At the beginning of each end step, each player sacrifices a permanent unless he or she discards a card." on the stack.
You get priority and pass.
Opponent gets priority and passes.
The top object on the stack (the ability put there in step 2B) resolves. You decline to discard a card, so you sacrifice Kynaios and Tiro of Meletis. I don't know what your opponent's decision is. This triggers the ability of Gift of Immortality.
You would get priority, but there is a state-based action to process. You put Gift of Immortality into your graveyard.
You would get priority, but there is a triggered ability to put on the stack. You put "When enchanted creature dies, return that card to the battlefield under its owner's control. Return Gift of Immortality to the battlefield attached to that creature at the beginning of the next end step." on the stack.
You get priority and pass.
Opponent gets priority and passes.
The top object on the stack (the ability put there in step 7) resolves. You return Kynaios and Tiro of Meletis to the battlefield. You set up a delayed triggered ability for the next end step.
You get priority and pass.
Opponent gets priority and passes.
The top object on the stack (the ability put there in step 2A) resolves. You draw a card. You don't draw a card. Each player has the option to place a land card from his or her hand onto the battlefield. Each opponent who declined that option draws a card does nothing.

This would enable you to use its ability again as you sacrifice Kynalos and Tiro of Meletis to the Possessed Portal ability. Note that outside that interval within the end step, the Gift of Immortality will be in the graveyard.

My question is, if I have the Gift attached to K and T... with an active Portal. Can i....
