I don't think it has to be Emrakul. It could very well be that Garruk, desperate for cure now, takes Vronos's advice and returns to Innistrad. Avacyn's solution could be to take the curse onto herself thinking she could handle it better than he could. Now instead of keeping the balance between humans, vampires and werewolves she kills all beings she comes across. This could lead Sorin to have to choose between trying to find a cure while she slaugthers the inhabitants of Innistrad or destroying his creation.

The three major Howlpack Alphas, the two remaining Vampire Lineage progenitors (Edgar Markov and Runo Stromkirk - Falkenrath's progenitor is dead), Ludevic (who is the only Innistrad character in a card name that doesn't have his own card yet), Jenrik, Volpaig (Avacynian Bishop and Leader of the Skirsdag cult).

So everyone has been focusing on the bloody feather and twisted symbol, but no love for the extremely prominent moon? With everyone asking for more werewolves, I would see some sort of Moon getting closer=more werewolves/ more aggressive werewolves. Would also be a nice way to work in Tamiyo, a popular planeswalker who has been left out in the dark for a long time.
I still think a day/night mechanic would be fun tho I guess the DFC Werwolves kinda does that already.
Imagine having cards that does something when it´s daytime on Innistrad and when Darkness Falls ***** is going to happend.

You can do more than just keywords, such as making opponents spells cost more on their turn (or yours), anthem effects, etc

I´m eager to find out if Innistrad will have Werewolves again or if there will only be Wolfirs. I just hope DFC returns since i loved the game aspect of that mechanic.
I'm pretty sure we will see werewolfs again, they were highly iconic in Innistrad. MaRo has stated that people really want a werewolf legend for Commander and I think they will deliver to this demand just as they did in OGW with the long awaited 5c Ally Commander.
Also, why should there NOT be Werewolfs in SOI? They are native to Innistrad, why would they be gone?
I can see Wizards doing away with DFC however, and I wouldn't bet on their return in SOI.
DFC's are extremely popular however, so there is no chance that WotC will do away with that mechanic. Granted, it won't show up in every block, but it will be used.

Seeing as how Shadows Over Innistrad will be the first set made under the 2 block system, I have some more faith in it, seeing how BFZ block turned out but was a set that got "damaged" by going from 3 sets to 2
