Here is is the basic minimum you can do to build your cube:
1) Balance your colors - typically for a 360 cube, 48 cards of each color, 50 artifacts, 30 multicolor and 40 lands.
2) Have a decent curve - Aggro colors must have decent 1-2 drops, etc. Check out other cubes to see typical curves.
3) Fixing is important - Do not scrimp on this - your drafters will thank you. Right now shocks are cheap - so buy them.
4) Choose a theme and size - be it pauper, graveyard, multicolor, artifact - or as with most cubes - the theme is simply powerful cards!
5) Don't worry about at first which cards are in and out. Use what you have now so you can play ASAP. You can then update your cube after a few games. As wtwlf123 said, a cube is never really complete, but conversely, a cube is always ready and playable!
