Posting this out of interest, as it seems to be somewhat of an emerging archetype. Multiplayer/EDH has gone from trash to treasure in Orzhov in the space of three months, thanks to Karlov and Ayli. Next to Vizkopa Guildmage (which was already insane with multiple players), this guild is now the business.
But there are more than likely enough lifegain cards out there in these colours that this becomes a valid archetype in all cube formats, and we're able to produce decks that hang together, gain life incidentally and then go ballistic with any of the key cards out. Blood Artist, Zulaport Cutthroat, Kokusho, BSA, SoLaS, Soul Warden and Loxodon Warhammer are all eminently cubeable in most formats. Is this solid enough to introduce yet? I know I'm close in Multiplayer...
As the owner of an EDH cube I'm considering putting in Lifegain again. Trostoni would love more support in that form and Karlov can replace one of the seven drop generals that there really shouldn't be two of. I know it's practically heretical, but I'd probably replace Vish Kal over Teysa 2. She's just a rock solid pillar to build control around and is an inevitable wincon. (Unblockable vigilance that can't take damage on the block; give that chick some credit!) Vish kinda fills the same niche as Ghost Council without being as fun and costing way more. I dunno, I kinda don't want to cut him completely but my Orzhov section is really tight and I'd want to put Vizkopa guildmage back in and maybe squeeze in Ayli.
This makes me want to rework a lot of Orzhov if I do this. Ugh. The big thing I have against lifegain is that Oloro is totally off limits because he does way too much passively and just sounds frustrating. So *the* big man in the archetype is off limits. Still Trostani andKarlov would still warrant a consideration.

The biggest problem with a lifegain subtheme at the moment is the massive lack of profit. By default the standard cube does provide enough gain - which can be pushed even more. But there aren't very much cards that enable effects due to lifegain like Archangel of Thune, Ajani's Pridemate and Karlov. And the latter both are more like filler cards than cards I'd center a strategy around.
Nevertheless my comment is not about Multiplayer cubes as I don't know practically how lifegain proves there ;-)
Yeah, there's probably more of a critical mass of cards there that have incidental lifegain. Kokusho, Sunscorch Regent and Wingmate Roc are all top-of-the-curve playables with incidental gobs of lifegain. Extort, too (e.g. Crypt Ghast, Pontiff of Blight). I suspect if you already find yourself running some sort of an Extort archetype in 1vs1 already, this might push it over the top, but I'm not sure if anyone's doing that at the moment.
